Holoprosencephaly is a severe genetic disease affecting human brain and craniofacial development. The work by Taniguchi et al. demonstrates that Tgif1, a gene implicated in human holoprosencephaly, is involved in regulating Sonic Hedgehog signaling and forebrain development during early mouse embryogenesis.
